The 41-year-old had decided to extend his international career and started the match in Lisbon at Estadio Jose Alvalade, where he began his professional career with Sporting 24 years ago.
Joao Felix scored the only goal in the 22nd minute, with his shot taking a slight deflection off Ben Davies.
Ronaldo missed a clear chance early in the second half and later had a goal ruled out for offside following a VAR check.
He was substituted as Portugal continued to create chances but failed to add to their lead.
Wales pushed for an equaliser late in the match, with Brennan Johnson hitting the crossbar before Ruben Dias made a crucial block from Lewis Koumas' follow-up effort.
Meanwhile, Erling Haaland scored twice as Norway beat Denmark 3-2.
In another match Germany and Netherlands drew 1-1.
